Abstract

The present disclosure provides a process for the preparation of crystalline polymorphic Form A of lacosamide.


Lacosamide (SPM 927, also referred to as Harkoseride or ADD 234037), is chemically (R)-2-acetamido-N-benzyl-3-methoxypropionamide.  

An article published in IP.com (Reference: IPCOM000187362D) discloses crystalline Form A of lacosamide.

Reported herein is a process for preparing Form A of lacosamide.  The XRD pattern of the prepared Form A is same as reported in the cited IP.com article.

            The process reported herein comprises the step of removing the solvent from a solution of lacosamide under vacuum to dryness to obtain crystalline Form A of lacosamide.  The preferred technique for the removal of the solvent is thin film evaporation, such as using a rotary evaporator. The solvent used herein is water, methanol, ethanol, dichloromethane, acetone, ethyl acetate, or mixtures thereof.

            The term “ambient temperature”, as used herein, is meant to indicate a temperature range of about 15°C to about 35°C.

EXAMPLES

Example 1:

Lacosamide (2 g) was dissolved in dichloromethane (20 ml) at ambient temperature and the solution was slowly (5 minutes to 10 minutes) transferred, under vacuum, to a preheated (95°C to 100°C) Buchi Rotavapor® flask.  The reaction mixture was maintained under vacuum at 95°C to 100°C for 10 minutes to 15 minutes and then cooled to ambient temperature. The resulting product was separated to obtain Form A.
