Preparation of hydrazine-free L-carnosine

L-carnosine has the following chemical structure:

Provided is a process for its preparation from (S)-N-(3-phthalimidopropionyl)histidine hydrochloride:

(S)-N-(3-phthalimidopropionyl)histidine hydrochloride was reacted with an excess of 40% aqueous methylamine using an alcohol as the reaction solvent. The reaction was performed under reflux and monitored by HPLC in order to complete the process.

The reaction mixture was cooled to the reflux temperature of an organic solvent that was added hot. The mixture was cooled to room temperature and filtered. The solid was purified by crystallization from water/alcohol in hot, filtered and dried in vacuum oven under heating to afford dry L-carnosine having more than 99% purity.
