N-(2-(4-(4-fluorobenzylcarbamoyl)-5-hydroxy-1-methyl-6-oxo-1,6- dihydropyrimidin-2-yl) propan-2-yl)-5-methyl-1,3,4-oxadiazole-2-carboxamide potassium salt (referred to as Compound 1) has the following chemical structure -

    Described herein below, is a process for preparation of 5-Methyl-1,3,4- oxadiazole-2-carboxylic acid potassium salt (referred to as compound 4). Compound 4 can be used as an intermediate for preparation of compound 1.

The process provides an efficient route for preparation of compound 4.

    Compound 2 can be prepared by reacting acetyl hydrazine with diethyl oxalate in the presence of inorganic or organic base (for example- triethylamine, diisopropylamine, diisopropylethylamine, diethylamine, pyridine).

The reaction can be performed with or without a solvent. Examples of suitable solvents are halogenated solvents, esters, hydrocarbons or ethers.

The process can be performed in suitable temperature in a range of from -25ºC to 70ºC. Stirring, may be done for a suitable time period, for example, from about 0.5 hours to about 78 hours.

Compound 2 can be obtained by the filtration .The work up involves optionally adding MTBE/toluene/ethyl acetate/Cyclohexane/heptane/Acetic acid to the reaction mixture.
