CRYSTALLINE FORM OF PALBOCICLIB

Palbociclib (codenamed PD-0332991, trade name Ibrance) is a potent and selective inhibitor of CDK4 and CDK6. It is chemically known as 6-acetyl-8-cyclopentyl-5-methyl-2-{[5-(piperazin-1-yl)pyridin- 2yl]amino}pyrido[2,3-d]pyrimidin-7(8H)-one, represented by formula-1.

Example 1: Preparation of Palbociclib form M1

Palbociclib (1 g) was dissolved in cyclohexanol (40 ml) at 130˚C. The clear solution was cooled to 25-30°C and stirred at same temperature for 24h. The solid obtained was filtered and dried at 40˚C under vacuum for 24h. The resulting product was identified as crystalline form M1 of Palbociclib. Yield= 0.7 g
